Part 3 – Submission of Fin Clip Samples


Permittees are required to submit fin clip samples for each broodstock used to produce juveniles for stocking in offshore systems under their Gulf Aquaculture Permit. Fin clip samples must be received at least 30 days prior to stocking juveniles in offshore aquaculture systems. For additional information or questions, please contact NMFS at XXX-XXX-XXX.


Follow these steps to obtain a fin clip sample:

  1. Clean a pair of stainless steel scissors with alcohol. Obtain a dime-sized sample of the fin by extracting it with the scissors and a pair of plastic forceps.1 Clean the scissors and forceps with alcohol between samples to minimize cross-contamination.

  2. Cut each sample in half and place each half into separate clean vials. Using a permanent marker, clearly label each vial with the following information: date sample was taken, tag number, sex, geographic location (e.g., GPS coordinates) where fish was collected, date when fish collected from wild, and the physical address where the fish will be held. If necessary, secure the writing area with scotch tape.

  3. Methods of preservation (please choose one):

    1. Deep Freezer - Immediately place the samples on ice or place in a deep freezer (-80oC).

    2. Ethanol preservation - Fill each vial with enough XX% non-denatured ethanol2 to cover the sample and place the sample in a deep freezer (-80oC).

    3. Dessication tubes – Place samples into dessication tubes and allow to dry for XX hours. After drying place the samples in a deep freezer (-80oC).

  4. Send one half of each sample along with the NMFS’ Fin Clip Log spreadsheet to NMFS. This spreadsheet and the NMFS address where samples will be received can be found at nmfs.sero.XXXX. Shipping should occur within 72 hours of harvest to preserve quality. Include a completed chain of custody form with each shipment. Contact NMFS at XXX-XXX-XXXX at least 24 hours prior to shipping to coordinate receipt of samples. Note that samples should be shipped early in the week to ensure that someone is available to receive the package. Samples should be packed with dry ice3 and shipped via next day delivery option.

  5. The aquaculture facility is required to maintain the other half of each sample as a back-up. These samples are to be kept in a deep freezer (-80oC) to preserve quality. Commercial grade freezers are not acceptable as they will lead to DNA degradation. Samples should not be disposed of without prior authorization from NOAA Fisheries.

The National Marine Fisheries Service requires this information for the conservation and management of marine fishery resources. 1 Remove dirt and any visible parasites from tissues as these can affect genetic analyses.

2 A license is required to purchase non-denatured ethanol as this is listed as a controlled substance.

3 Federal rules have been established which govern the shipment of dry ice. Please consult with your shipping company regarding any special instructions.
